What it is

SLU-PP-332 is a synthetic small molecule. The SLU is Saint Louis University. Thomas Burris, Cyrielle Billon, and colleagues built it to turn on ERR proteins — estrogen-related receptors. Those sit in the nucleus and switch on genes your muscle uses when you do aerobic work. They are not the estrogen receptor that people mean when they say “estrogen.”

It hits ERRα hardest, and also ERRβ and ERRγ. Shops call it an “exercise pill.” The papers say: mice on a treadmill lasted longer, and fat mice burned more fat.

Why it is not a peptide

No amino-acid chain. It is a small organic molecule. It is also not a steroid. It does not bind the androgen receptor. Putting it in the GH / sleep row is a category error. It does not release growth hormone the way CJC or MK-677 do.

What researchers studied

The 2023 ACS Chemical Biology paper is the introduction. Billon, Sitaula, Banerjee, Burris. In cells and in mice, SLU-PP-332 turned on the same gene program you see after a bout of aerobic exercise. Mice ran farther before they quit. When they knocked out ERRα in muscle, that extra endurance went away. So the effect they measured depends on that receptor.

The 2024 Journal of Pharmacology and Experimental Therapeutics paper put it in obese mice and ob/ob mice. Energy expenditure went up. Fatty-acid oxidation went up. Fat mass came down without them writing “they ate less” as the whole story.

There is no published randomized human trial that shows SLU-PP-332 does this in people. The molecule is a few years old. Anyone selling a human protocol is ahead of the literature.
